IIRC it can be used to detect that USB power is available, but AFAIK it isn't used in the bootloader.
Q2 and Q3 are just generic SOT-23 NPN transistors.
Siy has designed a neater solution with fewer components, and also provides ESD protection at https://github.com/siy/openstm32hw
but this requires SMD components.

One this though, it never asked me to install Serial driver for USB.... hm, but I was able to upload stuff to the board successfully, any ideas why it doesn't switch to SERIAL ?

When the Maple bootloader is uploading, it is a USB DFU device, so it doesn't need a serial driver to upload. What Operating System are you using? It may be that the Maples USB CDC serial is good enough to use the default driver; or you installed the driver ages ago?
The DISC pin asserts a USB disconnect by pulling the DP+ line low for a while. When it is up again, Windows searches for new devices on this USB port. If your board has a separate power supply, you could also unplug the USB cable, wait a second and replug it to have the same effect.
When no USB cable is plugged in (well, also when it is plugged in), the DP+ line must be high, pulled up by a 1.5k resistor. If it is low, no USB host will scan the bus for new devices. You can easily check that with a DMM.

I didnt get anything from ET Stamp. LED on PA5 keeps blinking no matter what I load into it! WTF?

Is there anything wrong here? Any idea why stuff I upload start?
Is my ET-STM32 is in perpetual bootloader mode ?
When I hit reset it blinks quickly 6 times, then keeps blinking with period of about 500ms. I can upload programs but looks like programs don't start. I've tried to upload in Flash mem or in RAM same effect. Hm. Plase advise.
If you are able to flash the boot loader over the serial port using the built in bootloader you are also able to use the serial port to read back the program you think you have flashed to 0x08005000 using USB. Just check if it is there.
If you see 1KB of 0xff, then 1KB of your program, the 1KB of 0xff, you flashed a bootloader for a low density device, but need one for a high density device, which the RET6 is. They differ in the size of the flash pages (1k/2k)
If you do not see it, flash it using the built in bootloader/serial port or install a proper boot loader.

I'm afraid I didn't catch what you mean... :(
I've uploaded maple_RET6_boot.bin downloaded from here. selected High density device 512K, flashed at 0x08000000.
